产品介绍： 阿拉丁生产的吉姆萨染色液(Giemsa Staining Solution)是一种用于染色体、细胞、血液、组织切片、细菌或原生动物寄生虫等样品染色的染色液。根据细胞成分的化学性质，吉姆萨染色液可将细胞质染成粉红色或蓝色，将细胞核染成紫红色。在光学显微镜下呈现出清晰的细胞图像，便于对细胞内部结构的观察及异常变化的识别。吉姆萨染色(Giemsa stain)，又称姬姆萨染色，是以德国化学家、细菌学家Gustav Giemsa的名字来命名的，最初是用于疟疾病人体内寄生虫的诊断，后由于对染色质和细胞核膜的高质量染色而用于组织病理学和细胞遗传学。吉姆萨染色液简单、省时、易于操作，对细胞质着色较好，结构显示清晰，但细胞核着色偏深，核结构显示不佳，故常与瑞氏染色液联合使用。阿拉丁的改良吉姆萨染色液、吉姆萨染色液和瑞氏染色液三种染色液的比较如下：吉姆萨染色液的主要成分是碱性染料天青II (azure Ⅱ)和酸性染料伊红(eosin)。天青II是一种碱性染料(阳离子染料)，常用于染色体或核酸的染色；伊红是酸性染料(阴离子染料)，是以氧杂蒽和醌式苯环作为生色基团，以羧基(-COOH)为助色基团的染料。染色的原理是基于在酸性染料中具有染色作用的阴离子和细胞内的碱性物质相结合，而碱性染料中具有染色作用的阳离子和细胞内的酸性物质相结合。各种血细胞成分的化学性质不同，对各种染料的亲和力也不一样。在对红细胞染色时，原始红细胞、早幼红细胞胞质、核仁含较多酸性物质，被染成较浓厚的蓝色；中幼红细胞既含酸性物质，又含碱性物质，被染成红蓝色或灰红色；完全成熟的红细胞，酸性物质彻底消失后，被染成粉红色。对于白细胞细胞质染色时，则分为以下几种情况：嗜酸性粒细胞中的嗜酸性颗粒呈碱性，与伊红结合，被染成粉红色；嗜碱性粒细胞中的嗜碱性颗粒呈酸性，与天青II结合，被染成紫蓝色；中性粒细胞中的中性颗粒呈中性，与伊红和天青II均可结合，被染成淡紫色；单核细胞或淋巴细胞的细胞质含有酸性蛋白质，被染成蓝紫色或紫红色。用于染色体染色即G显带时，染色体上富含A-T碱基对的DNA与组蛋白结合紧密，胰酶处理时不易被高度抽提，因而和染料结合力较强，呈深带，而富含G-C碱基对的区域结合的蛋白质被胰酶抽提，和染料亲和力降低，呈浅带。血细胞在本染色液染色后的细胞质颜色见下表：本产品染色效果好、染色力强、着色清晰。本产品用于HeLa细胞的染色效果参考图1。本产品以无水乙醇为溶剂，与传统的甲醇溶解法相比，提高了使用的安全性。按推荐稀释倍数配制染色工作液(1X)，且每样使用1ml染色工作液(1X)计算，本试剂盒的100ml和500ml包装分别可以检测1000个和5000个样品。室温避光保存，至少两年有效。血液涂片或骨髓涂片应厚薄均匀，以免影响染色和拍照。本染色液在稀释后应有类似金属光泽，否则染色液可能已失效。本染色工作液请用双蒸水、去离子水或超纯水等接近中性的水稀释，温度太低时偶尔会有少许沉淀，不影响染色效果。请切勿使用PBS稀释，否则会导致沉淀增多，影响染色效果。吉姆萨染色液对人体有毒性，且易燃。操作时请特别小心，并注意有效防护以避免直接接触人体或吸入体内。本产品仅限于专业人员的科学研究用，不得用于临床诊断或治疗，不得用于食品或药品，不得存放于普通住宅内。为了您的安全和健康，请穿实验服并戴一次性手套操作。使用说明： 1.试剂的准备工作吉姆萨染色工作液(1X)的配制：根据样品数量，取适量吉姆萨染色液(10X)，加入去离子水或双蒸水稀释10倍即成吉姆萨工作液(1X)。例如1ml吉姆萨染色液(10X)加入9ml水，混合均匀，即得10ml吉姆萨染色工作液(1X)。注：稀释后的吉姆萨工作液(1X)为即用型试剂，不宜保存，应现用现配，以免影响染色效果。2.样品处理a.对于石蜡切片：二甲苯中脱蜡5-10分钟。换用新鲜的二甲苯，再脱蜡5-10分钟。无水乙醇5分钟。90%乙醇2分钟。80%乙醇2分钟。70%乙醇2分钟。b.对于冰冻切片：蒸馏水洗涤2分钟。c.对于血液或骨髓涂片：按照常规方法制作血涂片或骨髓涂片，自然晾干。70%乙醇固定10分钟。d.对于培养细胞：加入70%的乙醇固定10分钟。3.改良吉姆萨染色a.切片或涂片样品加入适量配制好的吉姆萨染色工作液(1X)染色15-30分钟。注：如果染色过深或过浅应调整染色时间或工作液浓度。b.用蒸馏水从一侧充分洗涤，干燥后即可在显微镜下观察和拍照。Aladdin's Giemsa Staining Solution can be used to stain chromosomes, cells, blood, tissue sections, bacteria or protozoan parasites. The comparison of Aladdin's modified Giemsa Staining Solution, Giemsa Staining Solution and Wright's Staining Solution is as follows Precautions： Blood smear or bone marrow smear should be uniform in thickness for better staining and photos.This staining solution after dilution should have a metallic luster, otherwise it may be invalid.This product should be diluted with ddH2O, deionized water, or ultra-pure water. Do NOT use PBS for the dilution.
