脂溶性荧光探针促溶剂(Solubility Enhancer for Lipophilic Fluorescent Probes)是一种新型的高分子非离子型表面活性剂,常用于增强脂溶性荧光探针或染料的水溶性,提高荧光探针或染料的细胞通透性。本产品为500X的DMSO溶液,可直接用于细胞实验。脂溶性荧光探针如DiO (细胞膜绿色荧光探针) 、DiI (细胞膜红色荧光探针) 、DiD (细胞膜远红外荧光探针) 等,由于探针自身的溶解性问题,在配制染色工作液时可能出现沉淀,使用本脂溶性荧光探针促溶剂可有效解决沉淀问题,并能提高荧光探针的细胞通透性,使细胞膜染色更加快速,荧光染色更加明亮,染色背景更低。4℃保存,两年有效。-20℃可以保存更长时间。使用时请确保本产品已完全溶解。如果在溶解过程中有结晶析出,可在37-50℃加热以促溶解,不影响后续使用。本产品仅限于专业人员的科学研究用,不得用于临床诊断或治疗,不得用于食品或药品,不得存放于普通住宅内。为了您的安全和健康,请穿实验服并戴一次性手套操作。


使用说明

1.脂溶性荧光探针染色工作液中如果经常出现沉淀,可以尝试使用本产品。在配制染色工作液时,按照1:500额外加入本产品,通常就可以充分促进脂溶性荧光探针的溶解,并改善后续的染色效果。2.如下以DiO (细胞膜绿色荧光探针) 的配制为例进一步具体说明本产品的使用方法。其它的脂溶性荧光探针可以适当参考。a.细胞膜染色工作液的用量:对于6、12、24、96孔板,每孔的细胞膜染色工作液分别为1~2ml、0.5~1ml、300~500μl和50~100μl;对于流式细胞样品,每个样品的细胞膜染色工作液为0.5ml;对于切片,可以根据切片大小,每个切片使用100-200μl的细胞膜染色工作液。 b.DiO溶液的配制:DiO可以溶解于无水乙醇、DMSO和DMF,其中在DMSO溶解度大于10mg/ml (约11.3mM)。发现较难溶解时可以适当加热,并用超声处理以促进溶解。DiO的常用终浓度为1-30μM,最常用的终浓度为5-10μM。可根据实际情况配制成储存液,如储存液为5mM,实际使用时的终浓度为10μM,则此DiO储存液为500X。c.细胞膜染色工作液的配制:根据样品数量和每个样品所需工作液的体积,计算出细胞膜染色工作液的总体积。以流式细胞仪检测为例,每个样品的细胞膜染色工作液为0.5ml,参照下表配制细胞膜染色工作液。注意:请严格按照下表中组分顺序和体积配制细胞膜染色工作液,且DiO (500X)和Solubility Enhancer (500X)混匀后再加入染色缓冲液。染色缓冲液可以是正常的细胞培养液、无血清培养液、HBSS 或PBS ,或阿拉丁的细胞外液,本细胞外液可在一段时间内维持细胞的正常状态,并给细胞提供一定的营养,效果通常比PBS或HBSS更好,而使用正常的细胞培养液,可能会导致染色效果有所下降,此时需要适当考虑延长染色时间或加大染料浓度。 Reagent 2Sample 20Samples 200Samples DiO (500X) 2μl 20μl 200μl Solubility Enhancer(500X) 2μl 20μl 200μl AssayBuffer 996μl 9.96ml 99.6ml TotalVolume 1ml 10ml 100ml 注:不同细胞的最佳染色浓度略有不同,细胞膜染色工作液的最终浓度建议根据不同细胞系和实验体系进行优化。DiO的最终浓度提高时,促溶剂(Solubility Enhancer)的用量不变。d.按照DiO荧光探针的实际细胞染色方法进行染色。

Solubility Enhancer for Lipophilic Fluorescent Probes is a new type of polymeric nonionic surfactant, commonly used to enhance the water solubility of lipophilic fluorescent probes or dyes and improve their cell permeability. This product is 500X concentrated in DMSO and can be used directly in cell experiments.Lipophilic fluorescent probes, such as DiO (cell membrane green fluorescent probe) , DiI (cell membrane red fluorescent probe) and DiD (cell membrane far-infrared fluorescent probe) , may precipitate when preparing the staining working solution due to their low water solubility. This product can effectively solve the precipitation problem and improve the cell permeability of lipophilic fluorescent probes, leading to faster cell membrane staining with brighter fluorescence and lower staining background.


Precautions

This product must be completely dissolved before use. If crystal precipitates occur in this product, heat at 37-50℃ to promote dissolution, which has no effect on subsequent use. This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear a lab coat and disposable gloves during the operation.


Instructions for Use

1.Use this product to improve the water solubility of lipophilic fluorescent probes if precipitation frequently occurs in the their working solutions. The addition of this product into the working solution of lipophilic fluorescent probes at a ratio of 1:500 can effectively promote the dissolution of lipophilic fluorescent probes and improve their staining results.2.The following protocol is provided for the preparation of DiO working solution with this product. Preparation of other lipophilic fluorescent probes can be referred as appropriate. a.Amount of DiO working solution required: For per well of 6, 12, 24 and 96-well plates, the required amount of DiO working solution is 1~2ml, 0.5~1ml, 300~500μl and 50~100μl, respectively. For samples to be examined by flow cytometry, prepare 0.5ml of DiO working solution for each sample is. For sections, use 100-200μl of DiO working solution per section, depending on the size of sections. b.Preparation of DiO stock solution (500X): DiO can be dissolved in absolute ethanol, DMSO and DMF, with a solubility over 10 mg/ml (11.3 mM) in DMSO. Heat and ultrasonicate the solution appropriately when DiO is difficult to dissolve. A final working concentration of DiO at 5-10μM is most commonly used. DiO can be prepared into a stock solution as required. For example, prepare a 500X concentrated DiO stock solution (5mM) if a final working concentration at 10μM will be needed for staining. c.Preparation of DiO working solution: Prepare an appropriate amount of DiO working solution based on the number of samples. For samples to be examined by flow cytometry, each sample will require 0.5ml of DiO working solution, and prepare sufficient DiO working solution for different numbers of samples as follows: Reagent 2Sample 20Samples 200Samples DiO (500X) 2μl 20μl 200μl Solubility Enhancer(500X) 2μl 20μl 200μl AssayBuffer 996μl 9.96ml 99.6ml TotalVolume 1ml 10ml 100ml Note 1: DiO (500X) and Solubility Enhancer (500X) must be mixed well before adding the Assay Buffer. The Aassay Buffer can be normal cell culture medium, serum-free culture medium, HBSS , PBS , or 's Extracellular Solution . The Extracellular Solution can maintain cells in normal condition for a period of time by providing some nutrition to cells, and thus is usually better than PBS or HBSS. However, the use of normal cell culture medium may lead to a decrease in the staining effect, which can be avoided by extending the staining time or increasing the dye concentration appropriately.Note 2: The optimal staining concentration varies slightly from cell to cell, which should be optimized for different cell lines and experimental systems. When the final concentration of DiO is increased, the amount of Solubility Enhancer remains the same.d.Stain cells using an appropriate staining method with DiO fluorescent probe.Related Products:Cat.
